چکیده

SpoU is encoded by the fourth open reading frame (ORF) of the spo operon of Escherichia coli (1). The 5'-terminal gene of this operon, gmk, encodes guanylate kinase; the second gene, rpoZ, encodes the w subunit of RNA polymerase; the third gene, spoT, encodes ppGpp hydrolase, and the 3'-terminal gene, recG, encodes a helicase-like protein involved in recombination (1 and references therein). SpoU is the only functionally uncharacterized ORF in the spo operon. Comparison of the amino acid sequence of SpoU with amino acid and nucleotide sequence databases using the programs BLASTP and TBLASTN (2), respectively, revealed high similarity to an unannotated ORF from Rhodospirillum rubrum

SPOUT: a class of methyltransferases that includes spoU and trmD RNA methylase superfamilies, and novel superfamilies of predicted prokaryotic RNA methylases.

Nucleotide modification is a major source of structural and biochemical diversity of RNAs, particularly of rRNAs and tRNAs (Rozenski et al., 1999). Basespecific methylation is one of the prevalent modes of post-transcriptional modification along with pseudouridylation and thio-uridylation. The catalytic domains of the majority of RNA methylases belong to the vast la rge superc lass o f Rossmanf...

Novel plasmid-mediated 16S rRNA methylase, RmtC, found in a proteus mirabilis isolate demonstrating extraordinary high-level resistance against various aminoglycosides.

Proteus mirabilis ARS68, which demonstrated a very high level of resistance to various aminoglycosides, was isolated in 2003 from an inpatient in Japan. The aminoglycoside resistance of this strain could not be transferred to recipient strains Escherichia coli CSH-2 and E. coli HB101 by a general conjugation experiment, but E. coli DH5alpha was successfully transformed by electroporation with t...

Cellular Morphology and Immunologic Properties of Escherichia coli Treated With Antimicrobial Antisense Peptide Nucleic Acid

Background & Objectives: Antisense peptide nucleic acids (PNA) that target growth essential genes show potent bactericidal properties without cell lysis. We considered the possibility that whether PNA treatment influence the bacteria total nucleic acids content and apply approach to develop a new delivery system to Dendritic cells (DCs). DCs are the most potent antigen presenting cells in th...

Cloning and sequencing of ompf Salmonella typhi Salmonella ompf gene in Escherichia coli Origami

Background and Aim: Salmonella Typhi belongs to the family Enterobacteriaceae, gram-negative bacilli and causes gastrointestinal diseases such as typhoid. This bacterium has a special structure and various genes, including the ompf gene (outer membrane protein). Recent studies have shown the possibility of using ompf in the development of a diagnostic tuberculosis vaccine. Therefore, the aim of...

Enhanced Expression of Recombinant Activin A in Escherichia coli by Optimization of Induction Parameters

Activin A is a member of the transforming growth factor β super family. Because of its extensive clinical usages, its recombinant production is beneficial. In this study, activin A was expressed in E. coli using the pET 21a expression vector. The optimization of the activin A production in E. coli was done by using the response surface methodology (RSM). At this stage, the effect of IPTG and la...
